The principal objective of this chapter is to provide an overview of our current understanding of the structure and function of heme-containing peroxidases which belong to the plant and animal peroxidase superfamilies, with an emphasis on recent insights obtained from UV-vis electronic absorption and resonance Raman (RR) studies. In order to highlight the enzymatic mechanism of peroxidases, recent findings from multifrequency electron paramagnetic resonance (EPR) spectroscopy are also discussed. The relatively recently discovered family of bifunctional peroxidases, which are encoded by the katG gene in different bacteria, archebacteria and fungi, and which are phylogenetically related to heme peroxidases but are structurally and functionally more complex than the latter, are included.
