European tortoises of the genus Testudo are becoming seriously threatened mainly due to habitat urbanization and illicit pet trade. In this study we tested the crossamplifications of 23 microsatellite genetic markers (isolated from Testudinidae and Emydidae) in five (sub)species of the genus Testudo. A subset of 8–10 polymorphic loci was defined across the tested taxa, providing new tools for hybrid assignment, population genetic studies and parental tests.
