Abstract

The commercial use of lipases is a billion-dollar business that comprises a wide variety of different applications: these key enzymes find usage in a wide array of industrial applications, such as food technology, diary, chemical industry and biomedical sciences, paper refinement, nutraceutical, pharmaceutical, detergents. Psychrophilic lipases have lately attracted attention due to their low optimum temperature and high activity at very low temperatures, which are favourable properties in organic synthesis for the resolution of racemic mixture of relatively heat-labile compounds; in domestic cold-washing; in bioremediation applications carried out at ambient temperature (1-2). The recent availability of ten genome drafts from Antarctic marine bacteria belonging to the Pseudoalteromonas genus allowed us to embark in a data mining approach aimed at identifying gene encoding cold-adapted lipases. Several putative target genes were identified and some properties were predicted in silico. One of them has subsequently been chosen for recombinant heterologous expression in Escherichia coli. The recombinant cold adapted lipase accumulated in the insoluble fraction from which it was recovered in active form, purified to homogeneity and characterized.
