Gold nanoparticles (GNPs) decorated with glycans improve dendritic cells (DC) uptake, antigen-presentation and T-cells cross-talk. HIV-1-peptides loaded onto GNPs bearing high-mannoside (HM)-type oligosaccharides are presented to autologous T-cells from HIV-1 patients, increasing HIV-specific CD4+ and CD8+ T-cell proliferation and inducing highly functional cytokine secretion compared with HIV-peptides alone. Thus, co-delivery of HIV-1 antigens and HM by GNPs to dendritic cells enhances DC-presentation and improves highly functional HIV-specific cellular immune responses from HIV+ patients. This data would support the idea of combining GNPs bearing antigens and glyco-compounds as HIV vaccines.

Abstract

Gold nanoparticles (GNPs) decorated with glycans improve dendritic cells (DC) uptake, antigen-presentation and T-cells cross-talk. HIV-1-peptides loaded onto GNPs bearing high-mannoside (HM)-type oligosaccharides are presented to autologous T-cells from HIV-1 patients, increasing HIV-specific CD4+ and CD8+ T-cell proliferation and inducing highly functional cytokine secretion compared with HIV-peptides alone. Thus, co-delivery of HIV-1 antigens and HM by GNPs to dendritic cells enhances DC-presentation and improves highly functional HIV-specific cellular immune responses from HIV+ patients. This data would support the idea of combining GNPs bearing antigens and glyco-compounds as HIV vaccines.
