Abstract Successful clonal propagation of the ‘MKR1’ dwarfing persimmon rootstock (Diospyros kaki) via softwood cuttings has been previously achieved. However, in vitro techniques are essential to overcome seasonal limitations and ensure a consistent nursery stock supply. While in vitro propagation of Japanese persimmon has been established, utilizing dormant buds or single node sections as explants has been the norm. Nevertheless, varietal disparities necessitate tailored protocols for optimal results. Despite rooting experiments, no reports exist on the complete in vitro propagation of ‘MKR1’. This study aimed to determine optimal conditions for in vitro establishment, shoot proliferation, rooting, and acclimatization of ‘MKR1’.
