Side chain isotope labelling is a powerful tool to study protein structure and interactions. 1H,13C labelling of side-chain methyl groups in a deuterated background allows for extremely sensitive NMR experiments, useful to study large molecules that would be otherwise undetectable. On the other hand, side-chain aromatic groups are highly sensitive to the interactions with ligands, drugs, and other proteins. Moreover, such non-uniform labelling schemes greatly simplify the spectra of large proteins.
