This work explores the role of human GLRX3 in cytosolic [Fe-S] cluster assembly, focusing on its function in transferring [2Fe-2S] clusters to NUBP1 for [4Fe-4S] maturation. Using domain-specific constructs and mutants expressed in E. coli, we investigated GLRX3’s contribution through biochemical and spectroscopic approaches (UV-Vis, NMR, SEC).
