Cis-diamminedichloroplatinum (II) (cisplatin) is able to interact with human superoxide dismutase (hSOD1) with a dissociation constant in the mM range and with a slow kinetics. Cisplatin binds one cysteine (Cys) residue in each subunit of hSOD1 and/or only one Cys per dimer, in so weakening the quaternary structure of the enzyme. Cisplatin inhibits aggregation of demetallated hSOD1 in vitro and it is further able to dissolve and monomerize hSOD1 oligomers, thus indicating its potential as a drug for amyotrophic lateral sclerosis.
