|Abstract:||Soil microbes play a key role in maintaining soil health by carrying out various metabolic activities that are part of terrestrial biogeochemical cycles. Proteolysis is one such metabolic activity that is important for nitrogen mineralization. It is an essential component not only for terrestrial biogeochemical cycles but also for maintaining soil health and fertility,as it affects plant growth and soil biodiversity. To understand the link between microbial diversity and proteolytic potential, we are working to look at the distribution of soil microbes in different sizes of aggregates and at the same time the proteolytic potential present in these aggregates. For this purpose we have designed an experiment where we are using mirco, meso and macro aggregates of soil to look at the present microbial communities by looking at bacterial 16S ribosomal gene diversity. Moreover we will look at active populations after extracting RNA. This will enable us to to distinguish active populations in these aggregates. For measuring proteolytic potential of soil we will first measure the proteolytic enzyme activities in different aggregates, followed by study of proteolytic genes and their respective expressions. Using an advanced method we will try to measure extracellular and intra cellular proteases enzyme activity. By this study we expect to relate proteolysis with expressed genes. We expect that these studies will enhance our knowledge on selective distribution of bacterial populations in soil and proteolytic potential of soils based on different size of aggregates.|
