The purpose of this thesis is to obtain a tool for studying the connectomics of mouse and human brains through the combination of dierent techniques and the investigation of various clearing approaches. After studying the advantages and the disadvantages of various procedures present in literature, the goal was achieved through the development of a new optical clearing method for multi-modal brain imaging. This protocol is based on a water-soluble, versatile, simple, rapid and inexpensive agent: 2,2'-thiodiethanol (TDE)
