Glycation, is a nonenzymatic PTM of proteins in diabetic patients occurring between glucose and amino functions and resulting of an early stage Amadori rearrangement that leads to deoxyfructosylated lysine containing peptides and later to AGEs. Detection of early stage deoxyfructosylated peptides is difficult because of their low abundance in complex biological fluids. Therefore, development of efficient methods for enrichment and early detection is a challenge. Herein we report the novel stationary phase Boronic-ChemMatrix Resin (PhB-Lys(PhB)-CMRR) designed and optimized for an affinity chromatography to selectively capturing deoxyfructosylated peptides in different matrices. Its high selective affinity to deoxyfructosylated peptides vs differently glycosylated and unglycosylated peptides was demonstrated in an ad hoc mixture and in the hydrolysate of human serum albumin. Moreover we showed that the PhB-Lys(PhB)-CMRR is reusable after several cycles of capturing.
