This study investigates the role of the GLRX3-BOLA2 complex in the cytosolic assembly of [4Fe-4S] clusters and the maturation of NUBP1. Using a combination of spectroscopic and biochemical approaches, the researchers monitored Fe-S cluster formation and stability. UV–vis spectroscopy confirmed cluster integrity, while paramagnetic NMR provided direct evidence for [4Fe-4S] cluster formation on NUBP1. Anaerobic reconstitution assays further evaluated the efficiency of cluster assembly under controlled redox conditions. Together, these results shed light on the stepwise mechanism of [4Fe-4S] cluster biogenesis in human cells.
