The work on this thesis was focused on the understanding and elucidation of the structural and mechanistic properties of proteins involved in protein trapping and maturation in the intermembrane space of mitochondria, through the use of NMR spectroscopy. Three protein substrates of the Mia40/Erv1 machinery in the IMS were characterized: ALR, CHCHD5 and Tim9. Also the characterization of the interaction between two iron-sulfur proteins, Picot and Ciapin1, was performed. Finally, an E. Coli Zn and Pb detoxification ATPase, ZntA, was characterized.
