In this PhD thesis are reported the results on the development of a Surface Plasmon Resonance imaging (SPRi) based affinity DNA sensor for the ultrasensitive detection of human genomic DNA fragments of interest in molecular diagnostics. In particular the research activity is focused on the direct detection of sequences in unamplified target gene, by-passing the PCR amplification step. The whole strategy is developed around the MDR1 gene, taken as model for its potential relevance in theranostics and clinical analysis.
