This chapter will cover the main prodrug-related concepts, ranging from commonly accepted prodrug definition and classification criteria to the rationale behind prodrug design and development. A critical analysis of advantages and disadvantages associated with applications of drugs and prodrugs will be provided. The main objectives in prodrug design and development process, including pharmaceutical-, pharmacokinetic-, pharmacodynamic-related ones, with particular emphasis on permeability, solubility, metabolism, controlled release and targeting will be discussed.
